The process we want illustrated is a phenomenon called "endosomal release" of a small RNA molecule (siRNA) packaged in a lipid nanoparticle (LNP). We have attached a schematic of the process where a cell internalizes an LNP into an internal vesicle in the cell (an endosome) which is then damaged so that some of the siRNA in the LNP leaks out of the enclosing vesicle into the cytosol (we have included caps from a microscopy movie of this siRNA release). The damage to the endosome triggers the cellular cleaning system called the autophagy system which encapsulates the damaged endosome (with a double membrane). We attached a movie of this encapsulation of a damaged vesicle. We would primarily like to have this encapsulation illustrated (see red box in attached schematic) perhaps in with a 3D feeling similar to the attached example cover. One should see a damaged endosome with a stylized siRNA molecule leaking out and a half finished encapsulation of the endosome with autophagy membrane.
